Put in to bat first, Royal Challengers once again lost hard-hitting southpaw Chris Gayle (6) for a paltry score before de Villiers and Kohli crafted a 229-run stand for the second wicket, the highest partnership in the history of T20 cricket.
The Indo-South African duo also bettered their previous record of 215 not out against Mumbai Indians at the Wankhede last year.
Kohli (109) gradually accelerated his innings to secure his third century of this season, while De Villiers remained not out on 129, which included 12 sixes and 10 fours as Royal Challengers posted a daunting target of 249 for Lions.
However, Lions struggled to get a stable partnership as wickets kept tumbling in their attempt to match hovering run-rate. The visitors were eventually bundled out for 104 in 18.4 overs.
England fast bowler Chris Jordan was the pick of the bowlers for Royal Challengers, while spinners Yuzvendra Chahal and Sachin Baby took three and two wickets respectively.
Royal Challengers are still at sixth place, albeit with 10 points, above Kings XI Punjab and Pune Supergiants, while Lions are at second place despite enduring the record loss.
